Validation of a HS–GC–FID Method for the Quantification of Sevoflurane in the Blood, Urine, Brain and Lungs for Forensic Purposes

Background: Sevoflurane is a polyfluorinated compound extensively used as an inhalation anesthetic in patients undergoing surgery. If administered outside the operating room, sevoflurane dangerous and potentially lethal, toxicologists may be asked to investigate its presence biological matrices for forensic purposes. The aim of present study develop validate method detection quantification fluids organs through gas chromatography coupled flame ionization (GC–FID). Methods: was optimized based on maximization signal-to-noise ratio. GC–FID instrument equipped with Zebron capillary column ZB-624 (30 m, 0.32 mm ID, 1.80 µm film thickness). Results: validated over concentration range 1.0–304.0 µg/mL (blood urine) µg/g (brain, lungs). lower limit quantitation 1.0 or µg/g. Both intra- interassay imprecision inaccuracy were ≤15% at all quality control concentrations matrices. successfully applied measure 20 negative controls real case.
